Cohesin resolves to let go

چکیده
or microtubule asters, just as with life in general, sometimes you get stuck in a corner. And, say Cendrine Faivre-Moskalenko and Marileen Dogterom (FOM Institute for Atomic and Molecular Physics, Amsterdam, Netherlands), it can take a catastrophe to get things moving again. Dogterom has been trying to reproduce aster behavior in the minimal environment of a microfabricated chamber. Her beads, which are coated with cross-linked tubulin seeds to direct the growth of microtubules, are nudged to the chamber’s center as microtubules push on the chamber walls. But Dogterom realized that, in live cells, asters move and reposition constantly during mitosis. She mimicked these events by grabbing the beads with a laser trap. Beads placed in a corner stayed put, because the established, long microtubules bent around the chamber walls. But if Dogterom added Op18 protein to induce catastrophic microtubule shortening, the number of lengthy microtubules was reduced. Growing microtubules once again encountered the chamber walls head-on, allowing them to push the aster around. In the smaller cells of fission yeast, a microtubule that hits the cell surface might induce its own catastrophe. The force upon contact slows growth rate, and thus increases the likelihood that the microtubule will lose its protective cap. But in larger cells, the longer microtubules will buckle more readily. Here, peripheral motors may help pull asters to a central position. Dogterom hopes to test such a model by tethering motors to the walls of her chambers.

Chromosome Segregation: Learning to Let Go
To ensure accurate chromosome segregation, cohesion between sister chromatids must be released in a controlled manner during mitosis. A new study reveals how distinct centromere populations of the cohesin protector Sgo1 are regulated by microtubule attachments, cyclin-dependent kinases, and the kinetochore kinase Bub1.
متن کاملCdc14 Phosphatase Induces rDNA Condensation and Resolves Cohesin-Independent Cohesion during Budding Yeast Anaphase
At anaphase onset, the protease separase triggers chromosome segregation by cleaving the chromosomal cohesin complex. Here, we show that cohesin destruction in metaphase is sufficient for segregation of much of the budding yeast genome, but not of the long arm of chromosome XII that contains the rDNA repeats. rDNA in metaphase, unlike most other sequences, remains in an undercondensed and topol...
متن کاملScc2/Nipbl hops between chromosomal cohesin rings after loading
The cohesin complex mediates DNA-DNA interactions both between (sister chromatid cohesion) and within chromosomes (DNA looping). It has been suggested that intra-chromosome loops are generated by extrusion of DNAs through the lumen of cohesin's ring. Scc2 (Nipbl) stimulates cohesin's ABC-like ATPase and is essential for loading cohesin onto chromosomes. However, it is possible that the stimulat...
متن کاملPericentric Chromatin Is Organized into an Intramolecular Loop in Mitosis
BACKGROUND Cohesin proteins link sister chromatids and provide the basis for tension between bioriented sister chomatids in mitosis. Cohesin is concentrated at the centromere region of the chromosome despite the fact that sister centromeres can be separated by 800 nm in vivo. The function of cohesin at sites of separated DNA is unknown. RESULTS We provide evidence that the kinetochore promote...
متن کاملRoberts syndrome
All living organisms must go through cycles of replicating their genetic information and then dividing the copies between two new cells. This cyclical process, in cells from bacteria and human alike, requires a protein complex known as cohesin. Cohesin is a structural maintenance of chromosomes (SMC) complex. While bacteria have one form of this complex, yeast have several SMC complexes, and hu...
